Rival Livingstone took a seven-point halftime lead, and was led by Khyree Temple's 30 points.
But the Golden Bulls gave the packed home crowd a show in the second half, outscoring the Blue Bears 58-46 over the final 20 minutes.
JCSU's Ashton Sherrell scored 26 points, Avery Huggins and Javon Anderson poured in 21 apiece.
Now, Joyner can hit the 600 win milestone in Saturday's season finale at home against Claflin University.
